  "abstract" : [ "ger: Chlamydieninfektionen werden für eine Reihe von Fruchtbarkeitsstörungen und Atemwegserkrankungen beim Rind in Verbindung gebracht. Die entsprechende Literatur basiert in erster Linie auf serologischen Untersuchungen und weniger auf dem molekularbiologischen Direktnachweis der Erreger. Des Weiteren sind keine systematischen epidemiologische Studien zu finden. Demnach war das Ziel der Projektstufe I die Ermittlung der Erregerprävalenz in Milchviehbetrieben in NRW. Dafür wurden von den ca. 6.500 Milchviehbetrieben in NRW 100 Betriebe zufällig ausgewählt, wobei bei der Auswahl die Verteilung der Betriebe auf die Landkreise und die Anzahl Kühe pro Landkreis berücksichtigt wurden. Pro Betrieb wurden 10% der Herde (aber mindestens 10 Kühe) beprobt. Von den ausgewählten Kühen in der Frühlaktation wurden zwei Vaginaltupfer entnommen und diese zum einen bakteriologisch und zum anderen mit PCR-Nachweisverfahren untersucht. Die Proben wurden zunächst mittels real-time PCR genus-spezifisch (äußere Primer) auf Chlamydophhila spp. getestet. Im Falle positiver Nachweise dient das erhaltene PCR-Produkt als Matrix für die zweite Runde, die Spezies–spezifische PCR. Um die Prävalenz des Chlamydophila-Nachweises auf Landesebene zu ermitteln, wurde ein Betrieb als positiv eingestuft, sobald mindestens eine Kuh positiv getestet wurde. Diese Annahme ist durch die diskontinuierliche Ausscheidung von Chlamydien begründet. In den 32 Landkreisen des Landes wurden letztendlich von 100 milchviehhaltenden Betrieben insgesamt 1074 Kühe beprobt. Von den 1074 Tieren zeigte sich bei 145 Kühen (13,5 %) ein Chlamydophila-positives Ergebnis. Die Prävalenz auf Betriebsebene lag bei 61 %, d.h. in 61 % der Milchviehbetriebe in NRW war mindestens eine Kuh Chlamydophila positiv.
